The image represents the first time that five of the seven major mature tissue types of the highly regenerative planarian S. mediterranea are clearly visualized within a single image; each shown here with its own distinct color. According to this 10x single-cell RNA sequencing, virtually every dividing stem cell specifically expresses genes associated with one of the seven major tissue types. This finding significantly challenges how pluripotent stem cells are typically thought of. In contrast to the traditional view, what we see here supports a model of stem cell differentiation that does not involve “naïve” cells but rather involves a mechanism in which dividing stem cells that are already “specialized” toward a mature tissue type can switch their fate within the cell cycle.
